Negaplus
|
| Category : |
- NEGAPLUS contains Cefoperazone
- Cefoperazone is a semisynthetic third generation cephalosporin for
parenteral use
- Cefaperazone is a broad spectrum
antibiotic and active against many beta lactamase producing microorganisms
- Its activity against gram positive, gram negative and anaerobes makes
Cefaperazone an ideal empirical treatment
|
| Mechanism of action : |
- Cefixime is a beta lactam antibiotic.
It inhibits the cell wall synthesis of the bacteria, thus killing the
bacteria in the process.
- Detailed mechanism
of action
|
| Absorption : |
| Cefoperazone is not meant for oral use. The
drug is administered parenterally by IM or IV routes. |
| Higher peak serum levels are obtained when
the drug is administered as an IV bolus injection |
| Distribution : |
| Protein binding is about 82-93% |
| Cefoperazone is widely distributed in body
tissues and fluids. It must be noted that penetration of Cefoperazone into
the CSF is poor. |
| The drug crosses the placenta. It is also
excreted into breast milk. |
| Indication and usage : |
- Respiratory tract infections
- Intra-abdominal infections
- Septicaemia
- Bone and joint infections
- Pelvic inflammatory disease
- Urinary tract infections
|
| Contraindications: |
| Known allergy to Cephalosporins.
|
| Warnings : |
| In an individual allergic to beta lactam
antibiotics, the probability of allergy manifestation to Cefixime cannot
be ruled out. |
| Subsequent to the administration of Cefixime,
patients who present with diarrhoea should be considered for diagnosis of
pseudomembranous colitis. |
| Precautions : |
| In patients with severe biliary obstruction
or severe liver disease with renal disease, the daily dose of Cefoperazone
should not exceed 4 grams without close monitoring of serum levels |
| A disulfiram like reaction with flushing,
sweating, headache and tachycardia has occurred when alcohol was administered
within 24 hours or even upto 5 days after Cefoperazone injection. |
| Vitamin K deficiency has occurred in a few
patients. |
| It is important to monitor the Prothrombin
time when anticoagulants are used concomitantly. |
| Dosage: |
| 2-4grams/ day in 2 equally divided doses,
12 hours apart. In severe infections doses upto 16grams/day have been administered
without adverse side effects. |
| Paediatrics : |
| 50-200mg/Kg/day in 2 equally divided doses,
12 hours apart. Upto 300mg/kg/day has been used in severe infections including
meningitis. |
| Product formulation : |
| Cefoperazone for injection 1 gram |
| Benefits : |
| Negaplus has a broad spectrum and its activity
against Gram-positive, Gram-negative and anaerobic pathogens . This truly
broad spectrum and its activity even against beta lactamase producing strains
makes Negaplus suitable for emperical treatment of infections caused by
un identified pathogens. |
